A Novel Triplex Stable-Isotope Labeling Method for Quantitative Metabolome Analysis by LC-MS

摘要

An optimized derivatization method can offer higher labeling efficiency and high throughput for the triplex SIL reagents. LC-FTICR-MS has the capacity to quantify derivatized metabolites in the dynamic range of ~(12)C_(4)-:~(12)C_(2)~(13)C_(4)-:~(13)C_(4)- from 1:10:10 to 10:1:1. The results of relative quantification of 20 amino acids in healthy human urines from the 15-day/6-person project indicate very good reproducibility.
