This paper introduced a novel cell culture array chip with a concentration gradient-generating microfluidic structure. The chip was fabricated with poly (dimethylsiloxane) (PDMS) by the soft lithography. The Rhodamine solution was used to test the generation of the concentration gradients. The 3T3 cell was perfused into the microchannels and trapped in the dam array for culture. The Fluo-4AM solution formed concentration gradients via the microfluidic structure and then stained the cells respectively. The result verified that the chip succeeded in both the cell culturing and the generation of gradients, which was possible to apply for the drug screening and the system biology study.
