For the first time, how a variety of dietary fatty acids affect cellular acyl-CoA profiles was determined quantitatively. Hierarchical clustering in the remodeling of acyl-CoA profiles revealed a fatty acid-specific pattern across tested cell lines, which provides a valuable reference for making predictions in other cell models. C18:0 CoA and 18:2 CoA in the tumorigenic prostate cell line, C16:0 CoA, C18:3 CoA and C20:0 CoA in the tumorigenic hepatic cell line were altered differently by exogenous fatty acids, compared to the respective nontumorigenic cell line. This method provides a valuable tool for studying the implication of fatty acids and subsequent remodeling of the acyl-CoA profile, in the progression of tumors or other diseases associated with nutrition and/or metabolic abnormalities.
