Determination of Isoflavones in Different Parts of Trifolium Pratense by High-Performance Liquid Chromatography

An HPLC method was used for separation and determination of four isoflavone compounds (biochanin A, formononetin, genistein, and daidzein) from different parts (leaf, flower, and stem) of Trifolium pratense. The results showed that there were different distributions of the examined isoflavone compounds among different plant organs. The contents of total four isoflavone compounds in the leaf, stem and flower of trifolium pratense were 0. 856%, 0.403%, and 0.258% of dry matter, respectively, which was the highest in the leaf and the lowest in the flower. Biochanin A and formononetin were two predominant isoflavones in Trifolium pratense. The biochanin A content in different parts was 0.077% ~ 0. 358% of dry matter, which was the highest in the leaf and the lowest in stem. The formononetin content in different parts was 0.106% ~ 0.437% of dry matter, which was the highest in the leaf and the lowest in the flower. However, the amount of genistein and daidzein in different parts was low, whose content varied from 0.013% to 0.044% of dry matter.
