Al_2O_3 containing aw glass ceramic stimulates in vitro bone formation

Fresh marrow cells were obtained from femora of Fischer rats and cultured in a medium containing 15percent fetal calf serum (FCS) until confluence. After trypsinization, cells were subcultured at a cell density of 100 x 10~3/35mm well in the presence of FCS, beta -glycerophosphate, ascorbic acid phosphate and dexamethasone on four different culture substrata. The period of subculture was 2 weeks and the substrata used were culture dish, apatite-wollastonite containing glass ceramics (AW) having two different surface roughnesses, and Al_2O_3 containing AW (Al/AW). The glass matrix of AW contained MgO, CaO, P_2O_5 and SiO_2, whereas Al/AW contained Al_2O_3 in addition to these components. The subculture on all substrata showed many alkaline phosphatase (ALP) positive nodules. Though the DNA contents of the cells on these substrata were not significantly different, the number of nodules, ALP activity and bone Gla protein (BGP) mRNA level of the cells on Al/AW was the highest among these substrata. These results indicate that Al_2O_3 containing AW glass ceramic stimulates the osteoblastic differentiation of marrow stromal stem cells and therefore, the ceramic is useful material for the bone reconstruction surgery.
