Isolation and structural characterization of novel acid mucopolysaccharide from the viscera of Haliotis discus hannai

Two novel acid mucopolysaccharides were isolated from the viscera of Haliotis discus hannai, which were cultivated in Jiaonan county Shandong Province, by protease-hydrolysis combing CPC precipitated and purified to homogeneity by DEAE 52 and Sephacryl-S 200.The acid polysaccharides obtained, named as AVAP I and AVAP II, have an average molecular weight (MW) of 10.9 KD and 32.5 KD determined by HPLC coupled to a TSK-gel G4000PWXL column. And then their chemical characterizations were investigated. The results demonstrated that the polysaccharides were mostly composed of rhamnose, glucuronic acid, galactose with the approximate ratio of 3.69‶2.04‶1.0 and 6.40‶3.69‶1.0 respectively, besides including a small amount of xylose, fucose and mannose. The content of sulfate determined by ion Chromatography was 15.2% and 20.9% respectively and the content of protein was 4.44% and 4.12%.The structure characterization of AVAP II was evaluated by IR and 1H NMR, which verified monosaccharides composition of AVAP II. The configuration of rhamnose and galactose was α-pyranoid form, and the C-4 of some rhamnose was substituted by sulfate. These results suggest that the acid mucopolysaccharides from the viscera of Haliotis discus hannai are firstly reported, which possibly have some potential biological activity.
