Rapid propagation buds of Pinus massoniana in vitro

Using tender stems of the current-year in 1-year-old Pinus massoniana as explants and exploiting orthogonal experiments, we optimized the growth conditions of P. massoniana in vitro and successfully obtained the induction buds. The results were that: (1) as explants, tender stems that cultured in GD media supplemented with 2.0 mg/L N6-benzyladenine (6-BA) and 0.05 mg/L alpha-naphthyl acetic acid (NAA) have the best early differentiation rate of 100%; (2) among the tested conditions, GD modified media supplemented with 2.5 mg/L 6-BA, 0.03 mg/L NAA, total nitrongen content 350mg/L and 1∶2 of NH4+/NO3− is most suitable for propagation of adventitious buds giving a propagation rate and fold of 100% and 5.35, respectively; (3) GD modified media supplemented with 0.3% activated carbon is best for elongation of adventitious buds giving propagation rate of 485.9%; (4) induction period of buds were 40 day.
