Labeling of new formulation of tin-sucralfate freeze-dried kit withTechnetium-99m and it's biological evaluation

The present investigation deals with preparation of new formulation of tin-sucralfate freezedriedkit (F.D.K.), to be directly labeled with technetium-99m. The lyophilized form containing100mg sucralfate and 11.3mg dihydrated stannous chloride. The optimal pH values of the preparationwere found to be from 4.0 to 10.0. The range of sucralfate amount studied (50–500mg) not affectedthe radiochemical purity of the labeled complex. The radiochemical purity and the stability of thelabeled preparation that assessed by filtration were more than 95 %. Technetium–99m sucralfateradiochemical was stable up to a specific activity of 1000 mCi per gram which was more stable thanearlier published value, and without any radiolytic decomposition. The biological behavior of 99mTceluatewas evaluated in two groups of ulcerated and non-fasted mice. The data of organ distribution of99mTc-sucralfate in ulcerated fasted mice showed that more than 99 % of the administered dose wasaccumulated in the stomach (87.92 %) and intestine (11.43 %). The radioanalytical results togetherwith the in vivo-biological behavior of the labeled preparation demonstrate it's stability, efficacy andusefulness in medical applications for the detection of gastrointestinal ulcers.
