The objective of this study was to develop a biodegradable drug delivery system for the controlled delivery of ciprofloxacin HC1. The system (Atrigel~(~R)) consists of a biodegradable polymer dissolved in a biocompatible solvent. Various Atrigel~(~R) formulations with 10% ciprofloxacin HC1 in suspension were prepared using poly (DL-lactide-co-glycolide) (PLG and PLGH). Two formulations with 40% 50/50 PLGH (Ⅳ 0.2) or 75/25 PLG (Ⅳ 0.19) were selected for long-term release profile of ciprofloxacin HC1 in the rat. After subcutaneous injection in rats, implants and serum samples were collected at various time points over 28 days. Percentage of drug released from the implants demonstrated local sustained delivery of the antibiotic from the Atrigel~(~R) system for more than 3 weeks. The result shows that antibiotics can be delivered from this system for extended period of time after surgeries.
