Addition of an E8 Domain to Bone Morphogenic Protein 2 Confers Binding to HA Materials to Induce Osteoblastic Signaling

An octoglutamate (E8) was added to rBMP2 and this protein was successfully made through E. coli. rBMP2-E8 anchored tightly to HA materials in as short as 1 hour and stayed anchored even after 5 days of vigorous washing, unlike rBMP2. After heparin-affinity chromatography, purified rBMP2-E8 stimulated pSMAD 1/5 signaling in osteoblastic cells over untreated controls.
