These PNP hydrogels are exceedingly simple to prepare and easily scaled (requiring only simple mixing), and are highly biocompatible, containing components used extensively as biopharmaceutical excipients. We showed that the hydrogels enhanced the magnitude and duration of the humoral immune response compared to PBS delivery. We propose that the gel provides a local inflammatory niche as well as sustained release of the vaccine cargo. Together, these properties promote a prolonged germinal center response which enhance high affinity antibody production and humoral immune memory. This work introduces a simple and effective single injection vaccine delivery platform that increases the potency of subunit vaccines.
