The TA concentration that was most efficient at killing cancer cells without damaging healthy mammary cells was determined. Seeding TA crosslinked collagen type Ⅰ beads with human pre-adipocytes forms the basis for an injectable soft tissue regeneration matrix. As the adipocytes remodel the collagen the TA is released into the surrounding environment. When TA crosslinked beads of varying TA concentrations seeded with human pre-adipocytes were placed in a hydrogel then used to treat HER2~+ breast cancer cells the TA was capable of diffusing out of the matrix and inducing apoptosis in the breast cancer cells. Fig. 1 shows that an increase in TA concentration increases the cell death of HER2~+ breast cancer cells. Fig. 2 confirms that higher levels of TA in beads correlates to higher concentrations of TA in conditioned media. Because there is no difference in Bcl-2 between treatment groups and control, it indicates that the TA-induced apoptotic pathway does not affect the mitochondria. It appears the pathway is initiated due to endoplasmic reticulum stress.
