The use of supramolecular macrocycles to facilitate drug loading and delivery has a long history in the biomaterials field. However, routes to incorporate a precise number of nanostructures, and affinities between drug and macrocycle that are resistant to dissociation or competition, have remained elusive. Herein, we have generated an enabling platform using controlled growth dendrimer synthesis with high-affinity supramolecular macrocycles to create nanomedicine with modular and controlled drug loading. Our approach offers an additional level of therapeutic control in design of new nanomedicines.
