Conclusions: Using a novel semi-automated approach, the most significant effects on MSC morphology measured by quantitative cell shape descriptors were observed over time in 2D in low fibrinogen hydrogels. These changes in shape descriptors represent an elongated cell morphology (i.e. increased major axis length with increased aspect ratio, decreased roundness and increased circularity), consistent with a decrease in cell spreading and/or amount or extension of filopodia. This approach may be beneficial in targeted engineering of cell shape.
