Conclusion: This study is the first to demonstrate an in vitro 3D hydrogel model that upports the migration of NK cells, and demonstrates the selective interaction of NK cells with different cancer cell lines. Activation of NK cells has great potential for cancer immunotherapy, but understanding the mechanisms for tumor mediated inhibition of NK cell activity is critical. Three-dimensional tumor models that recapitulate cancer-immune cell interactions could be valuable tools to elucidating such mechanisms and advancing cancer immunotherapies.
