Three bone-targeting nanoprobes that varied in net charges and hydrophilicity/hydrophobicity were developed systematically by conjugating PAM on the backbone of different NIR fluorophores. The in vivo data suggest that net charges and hydrophobicity of targeted fluorophores play an important role on the biodistribution and clearance route as well as targetability and specificity. Thus the selection of a contrast agent will be important considerations in the development of next generation organ-specific imaging probes and therapeutic agents.
