Background. Post-surgical adhesion is an inevitable process in wound healing and results in many problems. A lot of biomaterials have been used to reduce the severity of adhesion. This study aimed to assess the effects of ibuprofen-loaded poly(E-caprolactone) (PCL-Ibu) film for acting as an anti-adhesion barrier.Methods. Ibuprofen was loaded to the PCL at different fraction and dissolved by ethyl acetate to fabricate various films respectively. The characteristics of PCL and PCL-Ibu films were evaluated by scanning electronic microscope (SEM), contact angle measurement, and in vitro bioadhesive strength test. Lactate dehydrogenase (LDH) test and Ibuprofen release assay were also investigated. Rat cecum abrasion model was used to evaluate the performance of Seprafilm, PCL film and PCL-Ibu(10) (PCL:ibuprofen=10∶1). Control groups did not receive any film treatment. Rats were sacrificed 7 days after surgery, and the severity of adhesion were investigated.Results. Contact angle measurement showed that hydrophilicity increased after ibuprofen loading to poly(e-caprolactone). Bioadhesive strength test revealed that ibuprofen loaded to PCL enhanced its adhesivity. Lactate dehydrogenase (LDH) test demonstrated minimal cytotoxicity for the PCL and PCL-Ibu(10) films. Ibuprofen release assay showed an initial burst release in the first 4 hours, followed by a plateau thereafter. After objective evaluation for areas of adhesion, the PCL-Ibu(10) group had smaller areas of adhesion, 2.4±7.59 mm~2. This was statistically significant lower than that of the Seprafilm (11.2±20.8 mm~2), PCL (9.8±16.5 mm~2), and control groups (50.4±25.3 mm~2).Conclusions. Both PCL and ibuprofen are not expensive and easily accessible. Ibuprofen-loaded PCL film improves the anti-inflammatory ability and provides a good anti-adhesion effect. The cost-effectiveness of Ibuprofen-loaded PCL film is quite promising in reducing post-surgical adhesion and should be regarded in health care system.
