Biomaterials designed to interact with cells are finding increasinguse in therapeutic delivery and regenerative medicine.These biomaterials have been employed for systemic administration, intended to deliver therapeutic agents throughout an organism, or local administration, where the agents are confined topathological sites. A cell interactive biomaterial allows one to deliver therapeutic agents directly into cells, rather than releasing the agents in the physiological milieu for diffusion and transport to target cells. Two approaches have been taken to impart cell affinity. In an approach intended to impart specificity, one relies on incorporation of specific ligands into biomaterials thatcorrespond todesirable receptors available on cellular surfaces. In an alternative, less-specific approach, hydrophobic (lipid)groups are incorporated into biomaterials, so that a natural affinity (or compatibility) to cellular membranes is imparted. Such amphiphilic biomaterials are expected to display solubility in the physiological milieu, which is critical fortherapeutic delivery, while displaying a broad cell affinity.This presentation will focus on development of amphiphilic biomaterials used in delivery of nucleic-acid based therapeutics.
