We examined the suppression of target gene using complex of oligonucleotides (ONs) and cationic liposomes (CLs) treated with high hydrostatic pressurization. The complexes of ON, which is designed to inhibit firefly luciferase expression, and CL were pressurized at various atmospheres (-10,000 atm), and were transfected to HeLa cells expressing firefly and renilla luciferase. The activity of firefly luciferase was effectively reduced by using the pressurized ON/CL complexes compared to the non-pressurized ON/CL complexes. Also, the selectivity of gene suppression was increased by using the pressurized ON/CL complexes. These results suggest the utility of pressure technology for modulation of gene expression by using oligonucleotides.
