Our investigation aimed to develop and test a nanofiber (NF)-collagen scaffold for the controlled release of bone morphogenetic protein 2 (BMP-2) for use as bone void filler, to stimulate new bone formation. The scaffolds were further enriched with platelet-rich plasma (PRP) extracts to provide an angiogenic stimulus emanating from the implanted biomaterial in the bone void.We tested our platelet-enriched NF-collagen sponge formulation by employing a rat surgical model of tibial bone "defect", where we temporally evaluated the controlled release of BMP-2 from the implanted scaffold using in vivo micro-Computed Tomography (micro-CT) to quantify micro-structural bone formation and graft integration, and to evaluate the revascularization and mineralization of the bone void defects.
