After separating the cap and stipe of Coprinus comatus, extraction method of protein in stipe was studied. Lipid was removed from stipe powder by soaking in ethanol, then protein was extracted with alkaline solution. The optimum extraction conditions deduced from response surface experiment are the following: stipe powder was extracted with 27 volumes of NaOH (pH 13) at 30 V for 0.5 h. For 100 g of stipe powder, 20.12 g of crude protein can be obtained. Its protein content was 41.05%, and polysaccharide was 19.67%. Using this method, protein in stipe can be effectively extracted while polysaccharide was less extracted. After extracting the protein of mushroom, the residue can be used to prepare the polysaccharide.
