Designing of Poncirus trifoliata Allele Identification Markers on All 9 Linkage Groups for CTV Resistant Citrus Breeding

Citrus tristeza virus (CTV) is an acute pathogen that is easily infected by aphid, and has caused serious damage to the Citrus industry. CTV resistant gene has been found in Poncirus trifoliata, which is a wild species related to Citrus. We have been performing to introduce the CTV resistant gene to Citrus cultivars from P. trifoliata by marker assisted back crossing. In this study, we developed CAPS markers that distinguish P. trifoliata alleles from Citrus alleles to identify the chromosome substitution in the backcross generation. Distinct alleles of P. trifoliata compared with any one Citrus species were observed in 42 markers. Twenty-six of 42 markers were mapped on the linkage groups constructed by the mapping populations. As a result, P. trifoliata allele identification markers were designed on all 9 linkage groups. This indicates that we will be able to breed CTV resistant Citrus by marker assisted back crossing.
