N-acetylcysteine (NAC) free acid caused significant plasticization and polymer instability when encapsulated in p'oly(lactic-co-glycolic acid) (PLGA) implants, resulting in high initial burst and short release duration of NAC at elevated drug loading (6-10 wt%). By encapsulating the Mg- and Ca-salt of NAC, the high initial burst, fast release, and accelerated PLGA degradation were each prevented, and excellent slow release was obtained.
