Accumulation of poly-beta-hydroxybutyrate (PHB) by photoautotrophic microorganisms makes it possible to reduce the high production cost. Effects of culture conditions on PHB biosynthesis in Synechocystis sp. PCC6803 cells have been studied in this paper, aiming to explore the way to enhance the PHB accumulation level in cyanobacteria. A method of PHB accumulation in cyanobacterium by two-step culture was carried out. Glucose and illumination promoted cell growth but did not favor for PHB synthesis. After 7 days of growth under nitrogne-starved photoautotrophic conditions, the intracellular level of PHB was up growth under nitrogen-starved photoautotrophic conditions, the intracellular level of PHB was up to 4.1
