Acetic acid was accumulated in fed-batch cultivation of Escherichia coli W3110 (pEC901) even though glucose was added slowly to keep the residual glucose concentration at an undetectable level. This was caused by the fluctuation of glucose concentration due to feeding and poor mixing at low agitation speed, and a high glucose concentration in the feed promoted acetic acid formation. When fed-batch cultures were conducted at a high agitation speed or by using a feed with relatively low concentration of glucose, the accumulation of acetic acid was greatly inhibited. Microfiltration was coupled to the fed-batch culture to remove accumulated acetic acid for 3 hours, and interferon activity increased by more than 4 times compared with that obtained in cultures without filtration.
