Improvement of Shikonin Production in Arnebia Euchroma Cell Suspension Culture Employing Two-Liquid-Phase Systems

A novel technology was developed with two-liquid-phase systems, which used an organic solvent to remove shikonin derivatives produced on cell surfaces during the culture of suspension cells of Arnebia euchroma. When n-hexadecane and oleic alcohol were added into the medium, the yields of shikonin derivatives were of 2.29 times and 2.45 times greater than that produced by the ordinary culture without solvent, respectively. The experimental results also indicated that the increasing volume of solvent could improve the amount of product excretion, but it would inhibit the growth of cells. When n-hexadecane was added on the 12th day of the culture, the maximal shikonin yield could be obtained, which was 2.49 times as much as that by the ordinary culture. Moreover, the effects of mixed solvents on shikonin production and cell metabolism had also been discussed in this paper.
