A Novel Separation Process for Stroma-Free Hemoglobin Production

With the concept of applying aqueous two-phase systems (ATPS) for separation and purification, a five-stage ATPS process, including micro fill ration, sonication, extraction, stripping and ultrafiltration units, was adopted to produce human stroma-free hemoglobin (SFHb). Not only were the performance of the units evaluated, but the correlation equations were also derived to model and optimize each unit operation. The process based on bench-scale equipment is capable of manufacturing 200 ml, 14 g/dl SFHb in 2.5 hour in a complete run. The function of SFHb in carrying oxygen is reserved and suitable for serving as the raw material of blood substitutes. The purity of SFHb obtained from the ATPS process was compared with the other three methods. The ATPS process is referred as to the most effective one in removing contaminated enzymes.
