A new kind of versatile linkers were prepared that permits both the covalent immobilization of prefabricated peptide and nucleic acids-such as oligonucleotides, PCR-products or peptide nucleic acid oligomers-and the in situ synthesis of such compounds on glass. Preparation of these linkers is very easy for researchers without chemical background by dipping way, and it is very flexible to change the length of linker, and flexible and hydrophilic property. The end function group can be easily changed to aldehyde, amino or hydroxyl group. Good repetition and uniformity fluorescent hybridizing images were achieved using ourselves developed protocol for spotting pre-fabricated oligonucleotides; In situ synthesis of DNA was successful using modified procedure. For both ways, an internal single base mismatch in 20mer oligonucleotides can be discerned from matched sequence. This set of linkers may be developed to fit almost all-current form biochips.
