The Damage Effects of Bisphenol A on Human Embryo Liver L-02 Cell Line

[Objective] To investigate the damage effects of bisphenol A on human embryo liver L-02 cell line and the mechanisms of it. [Methods] Human embryo liver L-02 cell were exposed to different concentration bisphenol A(1 μmol/L, 10 μmol/L,50 μmol/L, 100 μmol/L). Survival rate, level of SOD, CAT, GSH, MDA, apoptosis rate and the expression level of P53, caspase3 were measured. [Results] After exposed to bisphenol A, L-02 cell survival rate was proportional to the concentration of bisphenol A. Bisphenol A decreased SOD level significantly, GSH, MDA level decreased at 1μmol/L of bisphenol A and increased as the concentration of bisphenol A increased respectively. CAT level increased below 10 μmol/L of bisphenol A then increased significantly as the concentration of bisphenol A increased. Apoptosis and expression level of P53, caspase3 mRNA increased significantly at 50 μmol/L and 100 μnol/L bisphenol A respectively. [Conclusion] Bisphenol A have cytotoxicity effect and oxidative damage on L-02 cell line, induced L-02 cell line apoptosis also. Oxidative damage maybe one of the mechanisms of bisphenol A induced L-02 cell line apoptosis.
