摘要

Activaton tagging is a powerful means of isolating plant genes whose products are involved in complex biochemical processes. The dominant mutation produced allows direct selection for a defined phenotype. Plasmid rescue can be used to recover both the T-DNA and the flanking plant sequences containing the tagged gene. Activation tagging has been used to create a number of differing tobacco mutants, including those whose cells are characterized by their ability to grow in culture in the absence of auxin in the media. The tagged genes in this case are, in effect, cellular proto-oncogenes and are likely to play a role in the auxin biosynthetic and perception pathway.
