The radio-immunoassay and the enzyme linked immunoassay are the most commonly used method in field of analytical biochemistry. These techniques were performed on a plastic microtiter plate which was hydrophobically adsorbed antigen or antibody from solutions. The sequential reaction of immunostaining and occurrence of colour for detecting were performed on a microtiter. A new analytical method, thin-layer chromatography (TLC) has been developed in immunochemical assay field. It is called the overlay assay or TLC/enzyme immunostaining assay, This technique was used first by Magnani et al (1) in immunoassay for the detection of ganglioside GM1 on TLC plates using ~(125)I-labeled cholera toxin. After modification in various ways, this method has increasingly been applied to the field of the immunochemical assay. It has frequently been employed to detect carbohydrate structures of glycolipids with the specific ligands, such as anti-glycolipid antibodies, or lectins (2-3).. The application of the overlay assay has been extended to determine specificity and reactivity of virus or bacteria binding to specific receptor, and the enzyme hydrolyzing the substrate (4-5).
