To develop the non-invasive and effectively intradermal siRNA delivery system for treating atopic dermatitis, Tat as cell penetrating peptide and AT 1002 which reversibly increase paracellular transport of molecules across the mucosal epithelial barrier were used as intradermal delivery enhancers of siRNA. The fluorescence-labeled siRNA localization in mouse skin following the topical application of siRNA combined with Tat and AT 1002 analogs was observed by confocal laser microscopy, and the anti-allergy effects of siRNA for mouse RelA, which is NF-kB family, with Tat and AT 1002 analogs were evaluated in the allergic disease model mice.
