Variation in Ploidy of Anther-derived Regenerants in Brussels Sprouts (Brassica oleracea var. gemmifera)

Brussels sprouts (Brassica oleracea var. gemmifera) is one of a group of important coinmercial Brassica crops. Anther culture to create doubled-haploid lines is an important method for breeding of Regenerated plants from anther culture of Brussels sprouts are usually of mixed ploidy. However, the ploidy composition of such populations has not been reported by means of DNA flow cytometry. In this paper, variation in ploidy levels of four anther-derived regenerated populations was determined by DNA flow cytometry. Results showed that the four genotypes varied in their ploidy distribution. The newly bred F_1 hybrid EL5962 had the highest percentage of aneuploids. Two commercial F_1 hybrids, Maximus and Adagio, had similar ploidy distributions with the exception of no triploid in the regenerated population obtained from Adagio. The partial inbred line E327, gave the highest percentage of haploid and the lowest percentage of tetraploid and had neither triploid nor aneuploids. The percentage of diploid plants from three hybrids and one inbred line was very close, with the three F_1 hybrids being from 57% to 58% and the inbred line just a little lower at 54%. A comparison was made between our results and previous reports in Brassica oleracea species.
