Supercritical fluid chromatography (SFC) coupled to mass spectrometry is gaining in acceptance as a complementary technique to LC/MS. With SFC, one is able to achieve shorter analysis times, sharper chromatographic peaks and unique separations as compared to standard HPLC. SFC/MS has been applied in the high throughput combinatorial chemistry arena as well as with chiral separations. The next logical step was to investigate the use of SFC/MS for the analyses of potential drag candidates in animal plasma and serum samples, while keeping an eye toward rapid sample throughput. This aspect would prove to be a greater challenge since the biomatrix can be a much "dirtier" matrix than those noted from combinatorial chemistry type samples.
