    • 摘要: 运用植物基因工程手段构建琉璃苣BoD6D转化载体,为提高油料作物油份中γ-亚麻酸含量奠定基础.以琉璃苣基因组DNA为模板克隆BoD6D,构建酵母表达载体并转化酿酒酵母,对酵母进行诱导表达,提取脂肪酸后进行甲酯化反应,利用气相色谱分析脂肪酸含量;同时构建植物双元表达载体,经农杆菌介导通过蘸花法转化拟南芥,最后对转基因拟南芥通过抗性筛选及PCR进行鉴定.结果 表明,BoD6D编码区不舍有内含子,可以直接用于后续的功能研究;成功构建了酿酒酵母表达载体pYES2-BoD6D,气相色谱检测结果表明BoD6D在酵母中能够成功的将亚油酸催化生成γ-亚麻酸;成功构建了植物双元表达载体pBI121-BoD6D,卡那霉素抗性筛选和PCR鉴定结果表明已经成功获得BoD6D的拟南芥转化植株.通过以琉璃苣基因组DNA为模板比较方便的获得有功能的BoD6D用于转基因植物研究.

    • 摘要: 黑加仑籽油是从黑加仑籽中提取的一种新型营养保健油,富含γ-亚麻酸等多种不饱和脂肪酸和其他生物活性物质,且有多种保健功能,对人体健康有益.本文介绍了黑加仑籽油的主要成分、保健功效、提取方法、质量标准以及作为功能性油脂的发展潜力,以期为黑加仑籽油的开发利用提供理论基础.

    • 摘要: The culture medium and culture conditions of the mutant strain Mortierella ramanniana HLY0902 were optimized by orthogonal experiment and single factor experiment in order to increase the production of gamma-linolenic acid (GLA).The results showed that the yield of GLA was up to 1.05 g/L in the medium containing glucose 100 g/L,yeast extract powder 10 g/L,KH2PO4 4 g/L,NaNO3 1 g/L,MgSO4 · 7H2O 0.5 g/L and increased by 43.8% compared with that before optimization.The optimal culture conditions were identified as follows:inoculation amount 10%,loading amount 20%,batch time 168 h.Furthermore,the optimal conditions for mycelium growth and oil accumulation were pH 5.5 and 22°C,and the optimal conditions for GLA accumulation were pH 7.5 and 20 °C.The results showed that the capacity of GLA production by mutant strain Mortierella ramanniana HLY0902 improved through the optimization.%采用正交实验和单因素实验,分别对诱变菌拉曼被孢霉HLY0902的培养基及培养条件进行优化,以期提高γ-亚麻酸(GLA)的产量.结果表明:当发酵培养基组成为葡萄糖100 g/L、酵母浸粉10 g/L、KH2PO4 4 g/L、NaNO3 1g/L、MgSO4·7H2O 0.5 g/L时,GLA的产量最大,可达1.05g/L,较优化前提高了43.8%;最优培养条件为接种量10%,装液量20%,发酵时间168 h,适合菌体生长和油脂积累的pH为5.5、发酵温度为22°C,适合GLA积累的pH为7.5、发酵温度为20°C.通过该系列的优化研究,诱变菌拉曼被孢霉HLY0902产GLA的能力显著提高.

    • 摘要: 采用气相色谱法测定月见草子中γ-亚麻酸的含量.采用HP-INNOWAX毛细管色谱柱(40~260°C,30m×320μm×0.50μm)、FID检测器,载气为N2,纯度99.999%程序升温(初始温度150°C,保持5min然后以5°C/min速度升至240°C保持10min);分流进样进样(20:1);进样口温度为250°C;检测口温度为260°C,以外标法测定月见草子中γ-亚麻酸的含量.γ-亚麻酸在HP-INNOWAX毛细管色谱柱(40~260°C,30m×320μm×0.50μm)上具有较好的分离效果,其分离度为2.67.γ-亚麻酸的线性范围为1.2~7.2 mg/min(R2=0.9999);γ-亚麻酸平均回收率(n=9)为92.4%,RSD为0.4%.结论:所建立的方法简单、准确、可靠,可用于月见草子的质量控制.

    • 摘要: γ-亚麻酸(γ-Linolenic acid,GLA)是人体所必需且具有多种生理功能的一种重要多不饱和脂肪酸.研究表明,卷枝毛霉等GLA生产菌株中△6-脂肪酸脱饱和酶(△6-Fatty aciddesaturase,FAD6)是合成γ-亚麻酸的关键和限速酶,然而GLA是否惟一通过△6-脂肪酸脱饱和酶途径合成则目前还没有报道.作者以卷枝毛霉Mucor circinelloides EIM 10 sp.为研究对象,以其FAD6基因的5'端启动子和3'端终止子为同源臂,潮霉素B抗性基因为筛选标记,构建了M.circinelloides EIM10 sp.△6-脂肪酸脱饱和酶FAD6的敲除载体pUMCD6-HmB,转化进入制备好的原生质体中成功地敲除了FAD6基因,发现FAD6基因缺陷菌株GLA质量浓度比野生型对照菌株降低了50%以上,但GLA并没有完全消失,说明卷枝毛霉生物体内还有其他代谢途径可以将碳源转化为GLA.%γ-linolenic acid (GLA) that is an essential polyunsaturated fatty acid (PUFA) plays a vital role in many metabolic processes.Most related studies showed that delta 6-fatty acid desaturase (FAD6) was an important rate-limiting enzyme for the synthesis of γ-linolenic acid fiom Mucor circinelloides.However,it has not been reported whether the delta 6-fatty acid desaturase is the only pathway to produce γ-linolenic.In this work,we constructed M.circinelloides EIM10 sp.FAD6 knockout vector pUMCD6-HmB on the basis of pUC18 to study the function of FAD6.The homologous sequences of 5'promoter and 3'terminator of FAD combined with hygromycin B resistant gene used as the selective marker were connected as the knockout fragment.Then the vector pUMCD6-HmB was transformed into the prepared protoplast of M.circinelloides EIM10 sp.The results showed that FAD6 gene was successfully disrupted and GLA accumulation of the mutant disrupted in FAD6 decrease about 50% compared to the wild type.It can be concluded that FAD6 was essential for GLA production in M.circinelloides but there is probably other ways to accumulate GLA in M.Circinelloides.

    • 摘要: To optimize the Soxhlet extraction technology for borage see d oil, the influences of extraction time, ex-traction solvent volume and extraction temperature on the extraction rate of the borage seed oil were studied by single factor test and response surface method. The fatty acids in borage seed oil were determined by gas chromatography. The results showed that borage seed oil extraction rate reached the highest level (34.05%) under the conditions of extraction time 4.16 h, the volume of extraction solvent 108.81 ml and extraction temperature at 83.15°C. Totally 15 kinds of fatty acids were i-dentified in borage seed oil, including nine kinds of unsaturated fatty acids, among which, linoleic acid accounted for 37. 75%, gamma linolenic acid accounted for 20. 96%, and oleic acid accounted for 9. 20%.%本研究旨在优化索氏法提取琉璃苣籽油的工艺,以琉璃苣籽油提取率为指标,利用单因素试验和响应面法研究索氏法提取工艺中提取时间、提取剂体积和提取温度对提取率的影响,同时利用气相色谱仪对琉璃苣籽油中的脂肪酸进行分析鉴定.结果显示,粉碎的琉璃苣种子10 g,工艺条件为提取时间4.16 h,提取溶剂体积108.81 ml,提取温度83.15°C时,琉璃苣籽油提取率最高(34.05%).提取的琉璃苣籽油中共鉴定出15种脂肪酸,包括9种不饱和脂肪酸,其中亚油酸占37.75%,γ-亚麻酸占20.96%,油酸占9.20%.本试验的提取方法简单易操作,可为琉璃苣的产业化开发和利用提供依据.

    • 摘要: 为了获得深黄被孢霉合成γ-亚麻酸(GLA)的高产菌株,该研究采用了紫外照射和氮离子注入技术2种诱变方法建立突变体库.根据γ-亚麻酸代谢合成途径中的部分关键因子,设计并建立抗丙二酸和红四氮唑(TTC)筛选模型,旨在提高菌体生物量和不饱和脂肪酸含量.结果显示,采用丙二酸质量浓度为2g/L的平板可筛选得到高产油脂菌株;在发酵培养的第54小时收集菌体,用0.9%TTC染色4h,可作为TTC法的最佳染色条件.经过两轮不同方法的诱变筛选后,最终得到了一株具良好遗传稳定性的突变株I-0281,其GLA产量为889.80mg/L,比原始菌株提高了60.27%.

    • 摘要: 建立了-种毛细管气相色谱法,能同时测定黑豆中的α-亚麻酸和γ-亚麻酸.依次采用1%的硫酸-甲醇、15%三氟化硼-甲醇催化对提取出的黑豆脂肪酸进行甲酯化;以DB-WAX毛细管柱分离,FID检测器检测.结果表明:该柱对α-亚麻酸和γ-亚麻酸的分离效果良好,分离度>2,能完全分离;以十七烷酸甲酯为内标物,采用内标法测定α-亚麻酸和γ-亚麻酸的含量,其峰面积与质量浓度均有良好的线性关系,r>0.999 0;样品加标回收率(n=6)为87.5%~95%;RSD%分别为4.8%和0.9%,最低检出限为0.000 2 mg/mL.该方法能同时分离、测定黑豆脂肪酸中的α-亚麻酸和γ-亚麻酸.选取国内市场具有代表性的4种黑豆测定二者的含量,其范围分别为:α-亚麻酸:8 070 ~33 000 mg/kg;γ-亚麻酸:0~770 mg/kg.

    • 摘要: OBJECTIVE:To establish a method for the contents determination of phospholipid and γ-linolenic acid in Com-pound linolenic acid soft capsule. METHODS:HPLC was conducted to determine the content of soybean lecithin;the column was Agilent TC-C18 with mobile phase of methanol-water(84:16,V/V)at a flow rate of 1.0 ml/min,the detection wavelength was 219 nm,the column temperature was 30 °C,and the injection volume was 10 μl.Gas chromatography was conducted to determine the content of γ-linolenic acid in the preparation;the column was DB WAX,injector temperature was 210 °C by temperature pro-grammed,carrier gas was helium at a flow rate of 1 ml/min by split injection(split ratio of 1:30),and the injection volume was 0.02μl. RESULTS:The linear range was 4.64-16.24μg/ml for phospholipid(r=0.999 6)and 0.093 44-0.327 04μg/ml forγ-linole-nic acid methy ester(r=0.999 6);RSDs of precision,stability and reproducibility tests were lower than 3%;recoveries were 97.44%-99.36%(RSD=0.93%,n=6) and 97.22%-99.07%(RSD=1.01%,n=6),respectively. CONCLUSIONS:The method is simple,stable with good separation,and can be use for the contents determination of phospholipid and γ-linolenic acid in Com-pound linolenic acid soft capsule.%目的:建立测定复方亚麻酸软胶囊中大豆卵磷脂和γ-亚麻酸含量的方法.方法:采用高效液相色谱法测定制剂中大豆卵磷脂的含量:色谱柱为Agilent TC-C18,流动相为甲醇-水(84:16,V/V),流速为1.0 ml/min,检测波长为219 nm,柱温为30°C,进样量为10μl;采用气相色谱法测定制剂中γ-亚麻酸的含量:色谱柱为DB WAX柱,进样口温度为210°C,程序升温,载气为氦气,流速为1 ml/min,分流比为1:30,进样量为0.02μl.结果:L-α-卵磷脂和γ-亚麻酸甲酯检测质量浓度线性范围分别为4.64~16.24μg/ml、0.093 44~0.327 04μg/ml(r=0.999 9、0.999 8);精密度、稳定性、重复性试验的RSD<3%;加样回收率分别为96.79%~99.36%(RSD=0.93%,n=6)、96.63%~99.07%(RSD=1.01%,n=6).结论:该两种方法操作简便、稳定、重复性好,可用于复方亚麻酸软胶囊中大豆卵磷脂和γ-亚麻酸含量的测定.
