    • 摘要: Objective To investigate the effects of water extract of salvia miltiorrhiza aganst renal injury on rats exposed to cadmium.Methods Twenty female Wistar rats were randomly divided into blank control group (8),32 rats in model group,conventional breeding,the blank control group received intraperitoneal injection of saline 5 ml · kg-1 · d-1,model group with 2.8 mg · kg-1 · d-1 of cadmium chloride solution by intraperitoneal injection.After the success in modeling,the model group was divided into treatment group (salvia extract high dose 2.7 g · kg-1 · d-1,low water extract of salvia a dose of 1.35 g · kg-1 · d-1),negative control group,positive control group.After 4 weeks to experiment in the drug group,the water extract of salvia miltiorrhiza by intragastric administration,the positive control group using sodium selenite solution 0.05 mg · kg-1 · d-1 gavage,negative control group,the control group was given the same physical volume of saline water gastric 2.7 ml · kg-1.d-1,respectively.Each group intervention for 10 weeks.At the end of the experiment,serum was collected at,were used to detect the serum oxide dismutase (SOD),malondialdehyde (MDA),serum total antioxidant capacity (T-AOC).Results Danshen aqueous extract can make dye CD rats body weight growth faster,the serum SOD activity decreased significantly [(241.0 ± 2.8) U/ml vs (219.6 ± 4.6) U/ml,P < 0.05],serum MDA increased greatly [(7.2 ±0.3) μmol/L vs(11.9 ±0.9) μmol/L,P <0.01],to enhance the serum total antioxidant capacity [(8.7 ± 1.5) U/ml vs (4.6 ± 2.0) U/ml,P < 0.05].Conclusion Danshen aqueous extract has obvious antioxidant injury,in reducing swelling of renal tubular epithelial cells,improve renal function effect significantly.%目的 探讨丹参水提液对染镉大鼠肾损伤的抑制作用机制.方法 将雌雄各20只的Wistar大鼠随机分为空白对照组8只,模型组32只,常规饲养,空白对照组腹腔注射生理盐水5 ml·kg-1·d-1,模型组以2.8 mg·kg-1·d-1氯化镉溶液腹腔注射.待造模成功后,将模型组分为给药组(丹参水提液高剂量2.7g· kg-1 "d-1、丹参水提液低剂量1.35 g·kg-1 "d-1)、阴性对照组、阳性对照组.在实验进行到4周后,给药组采用丹参水提液进行灌胃,阳性对照组采用亚硒酸钠溶液0.05mg·kg-1·d-1灌胃,阴性对照组、空白对照组给予同体积生理盐水灌胃2.7 ml·kg-1 "d-1.各组干预治疗10周,于实验终末收集血清,分别检测血清血清超氧化物歧化酶(SOD)、丙二醛(MDA)、和总抗氧化能力(T-AOC)等指标.结果 丹参水提液可使染镉大鼠体重增长较快、血清SOD[(241.0±2.8) U/ml比(219.6 ±4.6) U/ml,P<0.05]明显升高、血清MDA[(7.2±0.3)μmol/L比(11.9±0.9) μmol/L,P<0.01]较大降低、能够增强血清T-AOC[(8.7±1.5) U/ml比(4.6±2.0)U/ml,P<0.05].结论 丹参水提液具有明显的抗氧化损伤的作用,在降低肾小管上皮细胞肿胀,改善肾功能方面疗效显著.

    • 摘要: 在病理条件下,机体内促氧化应激酶激活、线粒体呼吸链异常、氧自由基清除能力下降,导致氧自由基和促氧化因子生成增多.上述有害物质累积引起动脉管壁和血液成分发生相应的改变,如内皮依赖性舒张功能减退、炎性细胞因子激活、内皮结构破坏、血管平滑肌细胞增殖以及泡沫细胞形成等,从而在动脉粥样硬化的发生和发展过程中发挥着关键作用.目前,针对抗氧化治疗在心脑血管事件预防中的作用存在争议,有待进一步研究.%Under pathological conditions,promoting oxidative stress kinase to be activated in the body,mitochondrial respiratory chain abnormalities,and decreased oxygen free radical scavenging capacity lead to increased oxygen free radicals and pro-oxidant factors.The accunulation of the above harmful substances lead to the corresponding changes of arterial wall and blood components,such as endothelium-dependent relaxation dysfunction,inflammatory cytokine activation,endothelial structural damage,vascular smooth muscle cell proliferation,and foam cell formation,thus play key roles in the processes of the occurrence and development of atherosclerosis.The role of antioxidant therapy in the prevention of cardio-cerebrovascular events remains controversial and needs to be further studied.

    • 摘要: Idebenone is a synthetic analogue of coenzyme Q10. As a potent antioxidant,idebenone operates under low oxygen tension situations, protects cell membranes and mitochondria from oxidative damage through inhibiting lipid peroxidation, thereby protects against cerebral ischemia and nervous system damage. Idebenone also interacts with the mitochondrial electron transport chain and maintains the formation of ATP under ischemic conditions. Because idebenone has a good tolerability and safety, it is expected as a neuroprotective agent for the treatment of acute ischernic stroke. Studies in recent years have showed that idebenone has a certain therapeutic effect in a variety of nervous system diseases involving mitochondrial dysfunction and oxidative stress damage, such as mitochodrial encephalomyopathy, lactic acidosis, stroke-like episodes syndrome, Friedreich's ataxia, Alzheimer's diseases, Leber's hereditary optic neuropathy, and Duchenne muscular dystrophy. At present, some clinical trials in the mitochondria-related diseases and neuromuscular diseases are underway, and their results are expected to further expand the indications of idebenone.%艾地苯醌是辅酶Q10的一种合成类似物。作为一种强效抗氧化剂,艾地苯醌能在低氧张力状况下发挥作用,通过抑制脂质过氧化来保护细胞膜和线粒体免受氧化损伤,从而对抗脑缺血和中枢神经系统损伤。艾地苯醌还能与线粒体电子传递链相互作用,维持缺血状况下的ATP形成。由于艾地苯醌具有良好的耐受性和安全性,因此有望作为一种神经保护剂用于急性缺血性卒中的治疗。近年来的研究表明,艾地苯醌在各种涉及线粒体功能障碍和氧化应激损伤的神经系统疾病,如线粒体脑肌病、乳酸酸中毒和卒中样发作综合征、弗里德赖希共济失调、阿尔茨海默病、Leber遗传性视神经病变和Duchenne肌营养不良中均具有一定的治疗作用。目前,在线粒体相关疾病和神经肌肉病变中的一些临床试验JF在进行之中,其结果有望进一步拓宽艾地苯醌的适应证。

    • 摘要: 目的:为肿瘤的有效化学预防及新的治疗策略提供思路.方法:以H2O2与肿瘤发生、发展、凋亡的关系为基础,探讨了抗氧化药与过氧化氢(H2O2)的关系及其在肿瘤防治过程中的不同作用.结果与结论:肿瘤在新生、增殖、浸润、转移等各阶段都依赖于H2O2,因此可以通过药物改变细胞内H2O2的含量来影响其生长.对于具有体外抗氧化性药物的实际抗肿瘤作用,目前并未建立明确的评价策略,这就要求在使用这些药物时,必须注意使用剂量、对象并建立合理的体内评估机制.
