Industrial interest in potato proteins, which are produced as a by-product of potato starch processing, has increased due to the high nutritional value of these plant proteins. However, potato protein preparations may also contain the glycoalkaloids alpha-solanine and alpha-chaconine which, due to their toxicity, must be extracted. A method was therefore developed for extraction of these glycoalkaloids from potato protein powder under simulated industrial conditions, and their determination. The method involves acetic acid extraction/homogenization for removal of glycoalkaloids from potato protein preparations, solid phase extraction for concentration of the glycoalkaloids-containing extract and HPLC analysis using a Reprosil-Pur NH2 column. The method showed a limit of detection for both alpha-solanine and alpha-chaconine of 5 mug/ml; equivalent to 20 ppm in potato protein powder.
