目的:探讨 MALAT -1对胃癌细胞系 SGC7901侵袭和迁移能力的影响。方法:用实时定量聚合酶链反应(RT - PCR)的方法在 mRNA 水平测定 MALAT -1在20对胃癌组织及癌旁正常组织的表达,以及在 GES和 SGC7901细胞中的表达。RNA 干扰(RNAi)技术下调胃癌细胞系 SGC7901中 MALAT -1的表达后,以高内涵细胞运动试验检测其运动能力,Transwell 实验检测其侵袭和迁移能力。结果:RT - PCR 结果显示 MAL-AT -1在胃癌组织中的表达高于癌旁正常组织;相比 GES,MALAT -1在 SGC7901中的表达升高。下调 MAL-AT -1在 SGC7901中的表达后,高内涵细胞运动试验和 Transwell 实验结果显示 SGC7901的运动、侵袭和迁移能力均明显减弱。结论:MALAT -1能够促进胃癌细胞 SGC7901的侵袭和迁移能力。%Objective:To investigate the influence of MALAT - 1 on the invasion and migration in human gastric cancer cell line SGC7901. Methods:The mRNA levels of MALAT - 1 in 20 pairs of the gastric cancer tissues and ad-jacent normal tissues,GES and SGC7901 were determined by real - time PCR(RT - PCR)assays. The influence of MALAT - 1 on the mobility,invasion and migration of gastric cancer cell line SGC7901 were detected using high con-tent analysis and Transwell assay. Results:The results of RT - PCR showed that MALAT - 1 levels were up - regula-ted in gastric cancer tissues compared with adjacent normal tissues,and the expression level of MALAT - 1 in SGC7901 was significantly higher than that in GES. High content analysis and Transwell assay showed that the mo-bile,invasive and migratory ability were decreased after down - regulated the expression of MALAT - 1 in gastric cancer cell line SGC7901. Conclusion:MALAT - 1 could enhance the invasive and migratory ability of gastric cancer cell SGC7901.
