目的:探讨转染 let -7f -5p mimics/inhibitor 对胃癌细胞系 GC9811迁移和侵袭能力的影响。方法:用实时定量聚合酶链反应(RT -PCR)的方法在 mRNA 水平测定转染 let -7f -5p mimics/inhibitor 及其阴性对照后 let -7f -5p 表达水平的变化;以 Transwell 实验检测转染 let -7f -5p mimics/inhibitor 后迁移和侵袭能力有无变化。结果:RT -PCR 结果显示:转染 let -7f -5p mimics 可明显上调 GC9811细胞 let -7f -5p 的表达;转染 let -7f -5p inhibitor 可明显下调 GC9811细胞 let -7f -5p 的表达。Transwell 实验结果显示与对照组相比转染 let -7f -5p mimics 的 GC9811细胞迁移和侵袭能力明显减弱;与对照组相比转染 let -7f -5p inhibitor的 GC9811细胞迁移和侵袭能力明显增强。结论:microRNA let -7f -5p 能够抑制胃癌细胞 GC9811的迁移和侵袭能力。%Objective:To investigate the influence of microRNA let -7f -5p on the migration and invasion in hu-man gastric cancer cell line GC9811.Methods:The expression levels of microRNA let -7f -5p in GC9811 cell trans-fected with let -7f -5p mimics/inhibitor and its negative control was determined by Real -time polymerase chain re-action(RT -PCR).The influence of let -7f -5p on the migration and invasion of gastric cancer cell line GC9811 were detected by Transwell assay.Results:The expression of microRNA let -7f -5p was obviously up -regulated transfection of post let -7f -5p mimics and down -regulated by transfecting let -7f -5p inhibitor.Transwell assay showed that the migratory and invasive ability were decreased after down -regulated the expression of let -7f -5p and increased after up -regulated the expression of let -7f -5p.Conclusion:microRNA let -7f -5p could decrease the migratory and invasive ability of gastric cancer cell GC9811.
