miR-886-5p抑制肝细胞癌细胞侵袭与迁移

         

摘要

目的:探讨miR-886-5p在肝细胞癌(hepatocellular carcinoma,HCC)中的表达水平及其与临床病理特征的关系,并分析miR-886-5p对HCC细胞侵袭与迁移能力的影响.方法:应用Real-time PCR技术检测HCC组织和细胞系中miR-886-5p的表达水平;分析miR-886-5p的表达水平与HCC临床病理特征的关系;应用Transwell实验探究miR-886-5p对HCC细胞侵袭与迁移能力的影响.结果:miR-886-5p在HCC组织中的表达水平较癌旁组织明显降低(P<0.05),miR-886-5p在HCC细胞系中(Hep3B、MHCC-97L、HepG2、SMMC-7721)的表达水平较正常肝细胞LO2显著下调(P<0.05);miR-886-5p的表达水平与肿瘤数目、静脉侵犯、Edmondson病理分级及TNM分期显著相关(P<0.05);Transwell小室实验表明miR-886-5p能够显著抑制HCC细胞的侵袭与迁移能力.结论:miR-886-5p在HCC中表达下调,其能够抑制HCC细胞的侵袭与迁移.%Objective:To measure the expression level of miR-886-5p in hepatocellular carcinoma(HCC),analyze the correlations of clinicopathological features with miR-886-5p expression,and explore the effects of miR-886-5p on invasion and migration of HCC cells.Methods:Real-time PCR was conducted to detect miR-886-5p expression in HCC tissues and cell lines.The correlations of clinicopathological features with miR-886-5p expression were analyzed by statistical analysis.Transwell assay was applied to explore the functions of miR-886-5p in migration and invasion of HCC cells.Results:miR-886-5p was significantly decreased both in HCC tissues and cell lines(P < 0.05).miR-886-5p expression was remarkably associated with tumor nodules,vein invasion,Edmondson-Steiner grading and tumor-node-metastasis (TNM) stage (P < 0.05).Transwell assay revealed that miR-886-5p could inhibit invasion and migration of HCC cells.Conclusion:miR-886-5p was down-regulated in HCC,and could inhibit migration and invasion of HCC cells.
