To optimize the techniques,the response surface methodology and orthogonal design were employed to extract gastrodine from G.elata with the content as assess index.The results showed that un-der response surface methodology the optimum extraction techniques was as:enzymolysis temperature at 52.34 ℃,0.085 g of enzyme addition,as well as pH at 4.73.Under orthogonal design the optimum extrac-tion techniques was as:enzymolysis temperature at 55 ℃,0.09 g of enzyme addition,pH at 4.8.The ex-traction concentration of gastrodine reached 7.32 mg/g and 6.93 mg/g with response surface methodology and orthogonal design.%以天麻素含量为评价指标,用单因素方差分析法研究了酶解温度、酶解质量浓度和 pH 对天麻素提取含量的影响,并通过响应面法与正交设计法比较了天麻中活性物质的提取工艺和产率.结果表明:响应面法最佳优化工艺为:酶解温度为52.34℃,酶添加量为0.085 g 和 pH 为4.73;正交设计法的最佳优化工艺为:酶解温度为55.00℃,酶添加量为0.09 g和 pH 为4.80;通过响应面法酶解天麻后天麻素提取的最高含量为7.32 mg/g,而正交设计法天麻素提取的最高含量为6.93 mg/g.
