响应面法优化鱼骨中硫酸软骨素的醇沉工艺

O n the basis ofsingle factorexperim ent,response surface m ethodology w as used to optim ize alcoholprecipitation process ofchondroitin sulfate from fishbone.U sing sodium acetate concentration,pH value and ethanolconcentration asinvestigation factorsand chondroitin sulfate extrac-tion ratio as response value,the optim alpurification conditions w ere explored through a three-factor,three-levelcentralcom posite experim entalde-sign and the m ultiple quadratic regression equation w as acquired.R esults show ed thatthe optim um alcoholprecipitation process ofchondroitin sul-fate from fishbone w asasfollow s:sodium acetate concentration 4.0 g/100 m l,pH 6.0,ethanolconcentration 60% .The chondroitin sulfate extraction ratio w as1.910% .%在单因素试验基础上，采用响应面法优化鱼骨中硫酸软骨素的醇沉工艺参数。以乙酸钠质量浓度、pH值、乙醇体积分数为考察因素，硫酸软骨素得率为响应值，进行3因素3水平中心组合设计(CCD)试验，获得多元二次回归方程。结果表明，硫酸软骨素的最佳醇沉工艺为乙酸钠质量浓度4.0g/100mL、pH值为6.0、乙醇体积分数60%，硫酸软骨素实际提取率为1.910%。
