The present invention relates to a method and a device for detecting epigenetic modification of DNA in a sample by using plasmon coupling based on surface enhanced Raman spectroscopy (SERS) of immune gold colloids. According to the present invention, epigenetic modification of DNA (for example, circulating tumor DNA, and the like) existing in a sample at a very low concentration can be detected with high sensitivity and accuracy. Thus, the method can be applied to detect various biomarkers existing in body at a low concentration.
