UK-2 UK-2 UK-2 BIOSYNTHETIC GENES AND METHOD FOR IMPROVING UK-2 PRODUCTIVITY USING THE SAME

In order to provide a manufacturing method capable of mass-producing UK-2 at low cost, Strepto verticilum sp., which produces UK-2, to identify sites expected to be UK-2 biosynthetic gene clusters. 3-7 genomic DNAs were analyzed. Moreover, by colony hybridization, the DNA of this site was successfully isolated. In addition, the DNA was used to prepare a strain in which the gene present at the site was blocked. It has been found that this species does not produce UK-2. The genomic region was found to be a UK-2 biosynthetic gene cluster. In addition, streptoverticilum sp. 3-7 were transformed by the introduction of the vector into which the isolated UK-2 biosynthetic gene cluster was inserted. In addition, it was found that the productivity of UK-2 by the transformant was improved 10 to 60 times compared to the parent strain. Moreover, it was found that there were 2 copies of the UK-2 biosynthetic gene cluster per cell of each of these transformants.
