Cloning and analysis of GRF1 gene expressed in Moso bamboo during shoot elongation

We have cloned homologs to OsGRFl (Oryza sativa GROWTH REGULATING FACTOR1) gene from growing moso bambo shoot. There were three homologs in moso bamboo. PpGRFla had similarity to deepwater rice OsGRFl, PpGRFlc had similarity to japonica type rice OsGRFl and the rest one PpGRFlb had some deletion and insertion in investigated region. Southern hybridization analysis revealed that PpGRFl existed at least small multi gene family type in genome. Semi-quantitative RT-PCR analysis showed PpGRFl was highestexpressed at bamboo shoot. Also PpGRFl was expressed in growing young tissues, immature shoot apex and juvenile internodes and intercalary meristems. These results indicated that PpGRFl was growth regulating factor in moso bamboo and PpGRFl genes were constituted of small multi gene family. Interestingly, bamboo has both japonica and indica (deepwater rice) type OsGRFl. It might help us to consider the evolution of bamboo.
