Molecular Identification of a Novel PHB Producing Bacterium through 16S rRNA Sequencing

Polyhydroxybutyrate is a polyhydroxyalkonate, a polymer belonging to the polyester class. PHB accumulate as reserve energy material in the cytoplasm of certain bacteria during unbalanced growth conditions. As the production cost of PHB is quite high compared to non - degradable plastics, hence the present work has been focused on the isolation of potential bacterial strains from polluted water with high PHB accumulating ability. The PHB is produced by sodium hypochlorite method. The isolated strain shows high PHB producing ability (71%). Then the isolated strains are identified through both morphologically and genetically. Genetic level of identification was done through 16S rRNA. 16S rRNA genes were successfully amplified from the genome of the selected organism using universal primers. The partial sequences obtained were compared with sequences in NCBI data base using BLAST and the organism is identified as Bacillus cereus.
