METHOD FOR DETECTION OF MYCOBACTERIA BY TARGETING rRNA TO DETECT 16S rRNA DERIVED FROM SPECIFIC ACID-FAST BACTERIUM

PURPOSE: A method for detection of mycobacteria by targeting rRNA is provided. The method is useful for detecting 16S rRNA derived from a specific acid-fast bacterium with high specificity and sensitivity. CONSTITUTION: The method for detecting a specific acid-fast bacterium comprises using primers which have nucleotide sequences homologous or complementary to the specific regions in the rRNAs of the specific acid-fast bacterium to specifically amplify only the rRNAs of the specific acid-fast bacterium. An oligonucleotide for DNA elongation is provided, which binds to a specific region of the 16S rRNA of Mycobacterium tuberculosis and consists of at least 10 preferably consecutive nucleotides in either SEQ ID NO:1 or SEQ ID NO:2. An oligonucleotide for DNA elongation is provided, which binds to a specific region of the 16S rRNA of a nontuberculous acid-fast bacterium, Mycobacterium avium, and consists of at least 10 preferably consecutive nucleotides in either SEQ ID NO:3 or SEQ ID NO:4. An oligonucleotide for DNA elongation is provided, which binds to a specific region of the 16S rRNA of a nontuberculous acid-fast bacterium, Mycobacterium intracellulare, and consists of at least 10 preferably consecutive nucleotides in either SEQ ID NO:5 or SEQ ID NO:6. An oligonucleotide for DNA elongation is provided, which binds to a specific region of the 16S rRNA of a nontuberculous bacterium, Mycobacterium kansasii, and consists of at least 10 preferably consecutive nucleotides in either SEQ ID NO:7 or SEQ ID NO:8.
