摘要

FISH (Fluorescence In-Situ Hybridisation) is a technique used in the diagnosis of leukaemia to determine the disease phenotype, eg AML M1, M2 AML. Blood cells are grown in a short-term cell culture and a sample as a film is examined under a microscope to locate and identify areas exhibiting metaphase nuclei. Leukaemia type-specific probes are then pipetted onto these areas and a coverslip applied. After sealing of the covers-lips with natural rubber, the treated slides are then incubated in a hybridization oven and the fluorescence data obtained.
