Total Synthesis of (±)-Pentalenolactone A Methyl Ester

Pentalenolactone A (2) is a prominent member of a group of naturally occurring antibiotics (1-8 in Scheme 1) produced by prokaryotic organisms. These compounds exhibit a broad spectrum of activities against bacteria, fungi, viruses, and tumors. Biological evaluation of the pentalenolactones has demonstrated that they can specifically inhibit glyceralde-hyde-3-phosphate dehydrogenase in Trypanosoma brucei, thus indicating that these compounds could be potential candidates for the development of new antibiotic agents.
