摘要

CNS diseases such as glioma, stroke and multiple sclerosis are on the increase. However, many drugs are not being effectively and efficiently delivered to brain or CNS due to its structural complexity. The blood-brain barrier (BBB), characterized by semi-permeable multiple cell barriers with tight junctions, active enzymes and functional efflux transporters, is a major hindrance to drug delivery in the brain parenchyma [l]. Fewer than 2% of small-molecule drugs were able to penetrate into the cerebral parenchyma based on their excellent permeability through the BBB. Therefore, this is a huge hurdle in the development of therapies for CNS diseases, to maintain an effective concentration of drug in the brain.
